БА имеет многофакторную природу и развивается при тесном взаимодействии генетической предрасположенности и факторов окружающей среды. В последнее время большой интерес представляют исследования, посвященные участию врожденного иммунитета в развитии аллергических заболеваний системы. В различных популяциях мира показана ассоциация с развитием БА полиморфных вариантов генов Toll-подобных рецепторов. </p><p>Целью настоящей работы явился анализ ассоциации полиморфных локусов генов TLR1 и TLR6 с развитием БА у больных русской этнической принадлежности, проживающих на территории Республики Башкортостан. </p><sec><title>Материалы и методы</title><p>Материалы и методы. Материалом для исследования служили образцы ДНК больных БА неродственных индивидов (n = 179) и практически здоровых лиц (n = 121) русской этнической принадлежности. ДНК выделена методом фенольно-хлороформной экстракции. Генотипирование полиморфных вариантов осуществлялось методом полимеразной цепной реакции в реальном времени. </p></sec><sec><title>Результаты</title><p>Результаты. По результатам проведенного анализа выявлена ассоциация полиморфных локусов rs5743571 гена TLR1 и rs5743794 гена TLR6 с развитием БА. </p></sec><sec><title>Заключение</title><p>Заключение. Продемонстрирована значительная роль генов TLR1 и TLR6 в формировании предрасположенности к БА в русской популяции. Аллель rs5743571*C и генотип rs5743571*CC полиморфного локуса гена TLR1 являются маркерами повышенного риска, а генотип rs5743571*CT данного полиморфного варианта – маркером пониженного риска развития БА. Полученные данные будут способствовать лучшему пониманию механизмов защиты от атопической сенсибилизации и последующего развития атопической БА с участием TLR2-гетеродимеров.</p></sec></abstract><trans-abstract xml:lang="en"><p>The aim of this study was to analyze an association between TLR1 and TLR6 gene polymorphism and development of bronchial asthma in Russian patients living at the Republic of Bashkortostan. </p><sec><title>Methods</title><p>Methods. We investigated DNA samples of unrelated individuals (n = 179) and healthy volunteers (n = 121) of Russian ethnic group. DNA was extracted by phenol-chlorophorm extraction. Genotyping of polymorphic variant was performed by the real-time polymerase chain reaction. </p></sec><sec><title>Results</title><p>Results. An association between rs5743571 polymorphism in the TLR1 gene and rs5743794 polymorphism in the TLR6 gene with occurrence of bronchial asthma has been found. </p></sec><sec><title>Conclusion</title><p>Conclusion. TLR1 and TLR6 genes are important for asthma susceptibility in Russian population. Polymorphism in rs5743571*C allele and rs5743571*CC genotype of TLR1 gene could be used as high-risk markers; rs5743571*CT genotype of this polymorphic variant could be used as low-risk marker of asthma occurrence.
